Abstract
FIASCO is a high-efficiency method for constructing microsatellite-enriched genomic library. A microsatellite-enriched genomic library of Manglietia patungensis was constructed by using the method of fast isolation by AFLP sequences containing repeats (FIASCO). The microsatellite sequences and their characteristics were analyzed in this study. One hundred and nineteen clones which were 400 -500 bp in size were isolated and sequenced from Manglietia patungensis microsatellite libraries. Fifty seven clones of them contained microsatellites. Forty pairs of SSR-primers were designed and synthesized, among which 6 successful amplified the target regions,while none revealed microsatellite polymorphism. The influence factors for the low isolation efficiency of Manglietia patungensis were preliminary discussed.
