三尖杉科植物RAPD分析及其系统学意义

PHYLOGENETIC RELATIONSHIPS OF CEPHALOTAXACEAE BASED ON RANDOM AMPLIFIED POLYMORPHIC DNA

  • 摘要: 利用随机扩增多态性DNA(RAPD)技术分析了三尖杉科(Cephalotaxaceae)植物三尖杉Cephalotaxus fortunei Hook.f.、粗榧Cephalotaxus sinensis(Rehd.etWils.)Li、海南粗榧Cephalotaxus hainanensisLi和篦子三尖杉Cephalotaxus oliveriMast.,经筛选Operon公司的4组80个引物,其中14个引物的谱带清晰呈多态性。采用UPGMA法对各样本之间的遗传距离进行聚类分析,结果显示:在三尖杉科内建立篦子三尖杉组(Sect.Pectinatae)和三尖杉组(Sect.Cephalotaxus)是合理的。

     

    Abstract: Four species of Cephalotaxaceae were analyzed using random amplified polymorphic DNA(RAPD)markers.Eighty primers from 4 Operon kits were screened by RAPD,while 14 selected primers were able to produce clear polymorphic patterns.The results generated by using UPGMA algorithm to cluster genetic distance calculated from RAPD data imply that it is reasonable to establish the Sect. Cephalotaxus and Sect. Pectinatae in Cephalotaxaceae.
