附子内生菌及根际土壤性质与附子生物碱积累的关系研究

Relationships between endophytic structure, rhizosphere soil properties and aconite alkaloids accumulations in Aconitum carmichaelii Debx.

  • 摘要: 以不同产地附子及根际土壤为研究对象,采用高通量测序技术及高效液相色谱法对附子内生菌、土壤微生物、附子生物碱含量及土壤元素因子的变化规律进行了研究。结果显示,苯甲酰新乌头原碱、苯甲酰乌头原碱、单酯型生物碱含量与不动杆菌属(Acinetobacter)丰度、土壤pH值、Al3+含量与正相关,与曲霉属(Aspergillus)丰度负相关。次乌头碱含量与慢生根瘤菌属(Bradyrhizobium)丰度正相关,与苯甲酰次乌头原碱、次乌头碱含量与阿太菌属(Athelia)、镰刀菌属(Fusarium)的丰度负相关。研究结果表明,在田间管理中,增加土壤慢生根瘤菌属(Bradyrhizobium)、不动杆菌属(Acinetobacter)的比例,减少阿太菌属(Athelia)、镰刀菌属(Fusarium)、曲霉属(Aspergillus)的比例,并增加土壤pH值和Al3+含量可能提高附子中苯甲酰新乌头原碱、苯甲酰乌头原碱、苯甲酰次乌头原碱、次乌头碱和单酯型生物碱的含量。

     

    Abstract: In this study, Aconitum carmichaelii Debx. and rhizosphere soil from different areas were studied. The changes of endophytic microorganisms, soil microorganisms, alkaloid content and soil element factors of Aconitum carmichaelii were studied by illuminahiseq high-throughput sequencing technology and high performance liquid chromatography. The results showed that: the contents of benzoylneaconitine, benzoylaconitine and monoester alkaloids were positively correlated with Acinetobacter abundance, soil pH and Al3+ content,while negatively with Aspergillus abundance. The content of hypoaconitine was positively correlated with Bradyrhizobium abundance. However, the other two alkaloids (benzoylsubaconitine and hypaconitine) accumulations were negatively correlated with Athelia and Fusarium abundances. The results show that: changing the abundances of the key microorganism (such as Bradyrhizobium, Acinetobacter, Athelia, Fusarium and Aspergillus), and improving the Al3+ contents and pH in rhizosphere soil probably increase the contents of several alkaloids in Aconitum carmichaelii field management programs.
