  • The endosperm,dry leaves and fresh leaves of Larix chinensis were used as materials.By extracting DNA with an improved method of CTAB,digesting DNA with Mse I and EcoR I,pre-selective amplification,selective amplification,PAGE.FISH-AFLP technical system to detect genetic diversity in L.chinensis was established successfully.With primer combinations E-ACG/M-CAC and E-ACG/M-CAA,1856 and 2013 AFLP fragments were detected in during 28 individuals.Stable and abundant bands can be obtained with the materials of endosperms and fresh leaves.The techniques could be applied in the further analysis of genetic diversity in L.chinensis.
