Induction of Polyploid from Colchicine-Treated Fritillaria cirrhosa D.Don Callus

  • The callus of Fritillaria cirrhosa D.Don was obtained by placing the bulb on MS medium supplemented with 2 mg/L 6-BAand 1 mg/L NAAand 500 mg/L CH,and the callus was treated to get polyploid with colchicine.The result showed that polyploid callus was obtained by adding colchicine into the medium or by colchicine soaking.The former method was better than the later one.The best result for inducing tetraploid was by adding colchicine 1000 mg/Linto the medium treated for 5 days,the inducing rate was up to 70%.The chromosome number of the tetraploid was 2n=4x=48,while that of diploid was 2n=2x=24.
