DNA Extraction of the Endangered tree Species Cercidiphyllum japonicum Based on the Modified Method of SDS-CTAB

  • Six methods of DNA extraction of endangered tree species Cercidiphyllum japonicum are analyzed and compared in this article.The results showed that the modified method of SDS-CTAB is better than the others in terms of the quality of total DNA extraction.The value of A260/A280 is 1.8532 for DNA extracted by this of modified SDS-CTAB method.It’s higher than the others,such as 1.4872 for CTAB method,1.3552 for SDS method,1.5079 for PVP method,1.1858 fo1r urea method and 1.4534 for low pH medium with high salt method.The results of electrophoresis and PCR amplification also indicated that the modified method of SDS-CTAB is better than the others.
