Callus Induction and Establishment of Efficient Regeneration System of Arachis hypogaea L.

  • The tissue culture of peanut(Arachis hypogaea L.) was studied.The hypocotyls were used as explants on basal MS or 1/2 MS medium supplemented with different hormone combinations and agar.The result showed that the best medium for the inducement of callus was MS+6-BA 3.0 mg·L-1+NAA 0.4 mg·L-1+0.7% agar.Increasing of concentration of agar could inhibit vetrification and browning,and make callus easier to differentiate buds.The medium with 1/2 MS+NAA 1.0 mg·L-1+0.7% agar was favorable to the growth of roots and the rate of rooting was more than 95%.
